The rise of second-generation sequencing technologies revolutionized methodologies in human disease research.With the rapid development of high-throughput sequencing and analysis technologies，the data-driven，industrialized modern research mode is challenging the conventional hypothesis-driven mode，which significantly boost discoveries in disease-related genes and our understanding of diseases.Future widely applications of high-throughput sequencing technologies in"omics"would establish solid basis for individual genome annotation，disease prevention，diagnosis and personalized therapy.
